|
Effect of plasmid curing of Acinetobacter sp. B-W on theproduction of siderophore from glutamic acid as both carbonand nitrogen sole sources was investigated. Plasmid cured mutantof strain B-W lost the ability to produce siderophore fromglutamic acid at 28¡ÆC. Transformant E. coli DH5¥á harboring20 kb plasmid, that was isolated from wild type of strain B-Wproduced siderophore from glutamic acid as both carbon andnitrogen sole sources at 28¡ÆC, but, not at 36¡ÆC. Production ofsiderophore from glutamic acid by transformant E. coli DH5¥áwas completely inhibited by 10 ¥ìM FeCl3. In previous report,catechol nature of siderophore produced from glutamic acid bystrain B-W was detected by Arnow test. The siderophoreproduced from glutamic acid by transformant E. coli DH5¥áwas also catechol type. Rf value of siderophore produced fromtransformant E. coli DH5¥á grown in medium glutamic acid asboth carbon and nitrogen sole sources at 28¡ÆC was 0.32 inbutanol-acetic acid-water (12:3:5) as developing solvent. Rfvalue of the siderophore was the same with that of wild type ofstrain B-W. Thus a single plasmid of 20 kb seemed to beinvolved in the production of siderophore from glutamic acid.
